Research Interest
Dr. Huang’s lab has been working on deciphering the molecular pathogenesis of liver malignancy. Characterizing molecular pathogenesis of liver tumors will yield results toward identifying potential therapeutic targets for these deadly diseases.
Key areas of study include aspartate β-hydroxylase (ASPH), which catalyzes the hydroxylation of aspartate and asparagine residues within EGF-like domains, and ten-eleven translocation methylcytosine dioxygenase 1 (TET1), an enzyme responsible for generating 5-hydroxymethylcytosine and promoting DNA demethylation. Dr. Huang has demonstrated that ASPH is highly expressed in cholangiocarcinoma (CCA) but is minimally detectable in normal bile ducts.
Recent findings from his group indicate that several candidate compounds targeting ASPH enzymatic activity can effectively suppress CCA progression. Ongoing work aims to further define the therapeutic potential of these agents using multiple preclinical CCA models.
In parallel, Dr. Huang’s lab investigates TET1-mediated epigenetic regulation in the progression of alcohol-associated liver disease (ALD)–related hepatocellular carcinoma (HCC). These studies employ cell-type–specific TET1 knockout mouse models to dissect the role of TET1 in ALD/HCC pathogenesis. Since 2015, Dr. Huang’s research has been continuously funded by private foundations and federal agencies, including AASLD, NIH, and others.
